The Easiest Way to Get Them
 

A highly predictive siRNA design algorithm and stringent specificity checking are the keys to effective siRNA design. Ambion, through its partnership with Cenix BioScience, provides Silencer siRNAs — ready-to-use siRNAs designed for maximum potency and specificity using the most extensively tested siRNA design algorithm in the industry.

Although several web-based siRNA design tools are now available, almost none provide statistics as to the success rate of the underlying algorithm, as few have been rigorously tested. The few algorithms that have been analyzed have typically been tested using siRNAs to very few targets. Based on extensive tests performed at both Ambion and Cenix, it has become clear that small test sets do not accurately reflect the success rate of an algorithm, as some targets are more difficult to silence than others. The algorithm used to design Ambion's Silencer siRNAs has been functionally tested using >1100 siRNAs to nearly 400 endogenous human targets. This large data set (Figure below) shows that the algorithm successfully predicts effective siRNAs.
Silencer siRNAs Are Remarkably Effective. This graph shows the distribution of gene silencing measured for 808 siRNAs, designed using the Cenix algorithm, targeting >300 endogenously-expressed human genes. Target mRNA levels were measured by qRT-PCR, determining dCt values normalized against 18S rRNA from samples harvested 48hr after siRNA transfection into HeLa cells. These data demonstrate the effectiveness of the Cenix algorithm used to design Silencer Pre-designed and Validated siRNAs and Silencer siRNA Libraries. Data courtesy of Cenix BioScience.
