Experimental Workflow:
DNA Genotyping from Human FFPE Samples
Step 1. Isolate DNA from FFPE samples
DNA from formalin-fixed/paraffin-embedded (FFPE) tissues is characterized by fragmentation and chemical modifications.
Ambion's RecoverAll™ Total Nucleic Acid Isolation Kit helps overcome these limitations by maximizing the yield of DNA obtained from FFPE tissues.
Because DNA from FFPE tissues is heavily fragmented and chemically modified, only a small fraction is functional for genotyping. Determine how much is functional using TaqMan® RNase P Detection Reagents.
Step 3. Amplify DNA and collect allelic discrimination data
Sequence-specific forward and reverse primers and probes are used to amplify and detect the SNP of interest. The resulting allelic discrimination data is collected by performing an endpoint plate read on a Sequence Detection System such as the 7900HT Fast PCR System.
In this final step, a scatter plot is generated from the normalized allelic discrimination data and alleles are called to determine the genotype of each sample.
