mMESSAGE mMACHINE High Yield Capped RNA Transcription Kits utilize Ambion's patented high yield transcription technology to allow the routine synthesis of 15-35 µg of 7-methyl guanosine capped RNA from 1 µg of template DNA in a 20 µl volume during a 2 hour reaction. This is 10-50 times the yield of capped RNA obtained with conventional in vitro transcription reactions. The ratio of cap analog to GTP has been optimized to allow the best compromise between yield (15-35 µg) and proportion of transcripts that are capped (80%). The mMESSAGE mMACHINE kits also contain a LiCl precipitation solution that is efficient for separating proteins and unincorporated nucleotides (including free cap analog) from the capped RNA, allowing an increased efficiency of translation. Each mMESSAGE mMACHINE kit contains between 38 (SP6) and 57 (T7 and T3) A260 Units of cap analog - enough for 25 reactions. The cap analog:GTP ratio can be adjusted with the GTP stock solution provided in the kit to increase yields of long transcripts (>4 kb). The RNA synthesized from a mMESSAGE mMACHINE reaction is ideal for microinjection or in vitro translation studies.
In some situations, it may be necessary to completely remove the plasmid DNA template after RNA synthesis. For example, it is desirable to eliminate any residual DNA before using the nascent RNA as a standard for RT-PCR or in microinjection experiments. Ambion strongly recommends TURBO DNase for this task. As shown in the table at right, TURBO DNase is 63-fold more effective than standard DNase I in clearing DNA from in vitro transcription reactions, such as MEGAscript or mMessage mMachine, as measured by real-time RT-PCR.
NOTE: Each mMessage mMachine Kit contains 57 U of cap analog (SP6, 38 U). The mMESSAGE mMACHINE Kit is an outstanding value when you add in the cost of the Cap Analog.
mMESSAGE mMACHINE Kits are only optimized for use with the polymerases included in the kit. Using a different polymerase than what is supplied in the kit may result in low yields.
