Ambion's Decade™ Marker System produces a set of radiolabeled RNA molecules of 150, 100, 90, 80, 70, 60, 50, 40, 30, 20, and 10 nucleotides. The molecular weight markers are ideally suited for experiments where RNA molecules less than 150 nucleotides in length are being studied. As such, the Decade Marker System is particularly useful for research laboratories studying tRNA, snRNA, snoRNA, miRNA, siRNA and other small RNA molecules. It is an ideal companion for the MEGAshortscript Kit™, the mirVana™ miRNA Detection Kit, and the mirVana™ miRNA Probe Construction Kit.
The Decade Marker System includes all of the reagents necessary to produce the molecular weight markers except gamma-32P-ATP. The Decade Markers derive from a single, gel-purified transcript that is provided. The transcript is 5' end-labeled using the kinase and kinase buffer supplied with the kit. The kinase reaction is then diluted into a cleavage reagent that generates the molecular weight marker set in a five minute, room temperature reaction. Gel loading buffer from the kit is then added to the Decade Markers to ready them for electrophoresis. The labeling, cleavage, and gel preparation steps can be completed in less than an hour. Depending on how much radioactivity you require for your experiments, Decade Markers from a single reaction can be used for 5-20 experiments.
There are enough Decade Marker and accessory reagents for ten preparative reactions, providing RNA markers for as many as 200 experiments.
