A successful siRNA experiment requires the appropriate transfection agent for your cells, and one that is optimized for siRNA delivery. The efficiency with which different transfection reagents deliver nucleic acids can vary dramatically with the type of nucleic acid being transfected (RNA vs. DNA, single-stranded vs. double-stranded) and the cells being used.
Ambion has developed transfection agents optimized for the transfection of siRNA into a wide variety of cultured cells: siPORT Amine Transfection Agent (a polyamine mixture) and siPORT™ NeoFX™ Transfection Agent (a cationic and neutral lipid mixture). As expected, the two agents have different transfection efficiencies for different cell types, but together allow siRNA to be introduced into a broad variety of cultured cell types. Furthermore, siPORT Amine and siPORT NeoFX can be used to transfect cells during subculturing—saving a full day of valuable time—without increased cytotoxicity.
