The BrightStar BioDetect Kit is a low background, high sensitivity, nonisotopic detection system compatible with Northern, Southern, dot blot and nuclease protection assays. It contains all of the reagents and materials necessary for detection of biotinylated RNA and DNA probes. We strongly recommend BrightStar-Plus Membranes with this system, as they have been optimized to work together. BrightStar-Plus Membranes are available separately in a selection of sizes. When used together, these components yield results with exceptionally high sensitivity and low background. BrightStar BioDetect also offers the advantage of one of the brightest and longest-lived chemiluminescent substrates, CDP-Star™, providing maximum sensitivity and the option of multiple exposures over a two day timeframe.
Nonisotopic Northern Analysis
Many researchers have been frustrated with their attempts to perform nonisotopic Northerns because of the high background and low sensitivity associated with this technique. The sensitivity of the chemiluminescent procedure is limited by a typically low signal-to-noise ratio, which is attributable to two sources - nonspecific binding of the probe and the secondary nonisotopic detection component to the membrane. The BrightStar BioDetect Nonisotopic Detection System was developed in conjunction with Ambion's NorthernMax® Kit to eliminate these sources of background. BioDetect utilizes an optimized membrane, blocking and wash solutions, and protocol that provides
an extremely sensitive assay. All solutions supplied are ready-to-use, making the chemiluminescent procedure simple, reproducible and convenient.
Nonisotopic Ribonuclease Protection
A ribonuclease protection assay (RPA) format can also be used with the BrightStar System, providing additional sensitivity in mRNA detection. In an RPA, the hybridization of a biotinylated antisense RNA probe to total or poly(A) RNA is carried out in solution rather than on a membrane, as in Northern analysis. This eliminates one of the major sources of background signal seen with Northern analysis. When used in conjunction with Ambion's HybSpeed™ High Speed Ribonuclease Protection Assay Kit or RPA III™ Kit, BrightStar BioDetect provides a highly sensitive chemiluminescent mRNA detection system with greater sensitivity than a standard 32P-Northern assay.
The BioDetect Kit provides all the necessary buffers, streptavidin alkaline phosphatase conjugate and chemiluminescent reagent sufficient for more than 1000 cm2 of membrane.
