The RNAqueous-4PCR Kit provides RNA that is free of genomic DNA contamination. The kit includes reagents to remove contaminating DNA from the isolated RNA, making this kit especially suitable for use in conjunction with RT-PCR applications. The DNase treatment and removal reagents consist of RNase-free DNase I, 10X DNase Buffer, and a DNase Removal Reagent. In addition, the kit contains three plastic pestles designed for disruption of small amounts of tissue.
More About Sample Size
The RNAqueous-4PCR Kit is designed for RNA isolation from small tissue samples (0.5-75 mg) and small- to medium-sized cultured cell samples (~100–107). These sample size ranges overlap with the capabilities of the both the RNAqueous and RNAqueous-Micro Kits. For samples across this entire size range, choose the RNAqueous-4PCR Kit to isolate RNA that will be used in applications, such as RT-PCR, that would benefit from its robust genomic DNA removal capabilities. For samples at the lower ends of these sample size ranges, choose either the RNAqueous-Micro Kit or the RNAqueous-4PCR Kit based on the size of the majority of your experimental samples.
A Robust Procedure
The RNAqueous-4PCR Kit utilizes Ambion's RNAqueous technology of binding total RNA to glass fiber filters in a microfuge tube format. Briefly, the procedure consists of disrupting tissues or cells in a guanidinium-based lysis solution, followed by binding of the RNA to a glass fiber filter, washing the filter to remove contaminants, and then eluting the RNA in a small volume (at least 50 µl) of elution solution. The kit also contains linear acrylamide, a coprecipitant that may be used for concentrating the eluted RNA by alcohol precipitation.
When used in conjunction with Ambion's RNAlater® Tissue Collection: RNA Stabilization Solution, most tissue samples can be stored at 4°C and then easily disrupted in standard microfuge tubes using the pestles provided in the kit. There is no need for snap-freezing in liquid nitrogen or grinding in a mortar and pestle.
The RNAqueous-4PCR Kit includes enough reagents for 30 RNA purifications.
