The RNAqueous Kit is designed for the purification of total RNA from small-scale samples. The amount of tissue that can be used in a single RNAqueous prep varies depending on the tissue source; generally 1 to 75 mg of tissue or 100 to 107 cultured cells is an appropriate amount of starting material. In the procedure, RNA is bound to a glass fiber filter housed in a microfuge tube fitted assembly. Solutions are driven through the filter by microcentrifugation or by using a vacuum manifold. The RNAqueous Kit contains enough reagents for 50 RNA purifications.
RNAqueous Technology RNAqueous kits can be used to purify total RNA from many different tissues and cells in a fast procedure without the use of phenol. Cells or tissue are disrupted in a guanidinium thiocyanate solution; this chaotropic agent effectively lyses cells and inactivates endogenous ribonucleases. The lysate is then diluted with an ethanol solution and applied to an RNA-binding glass fiber filter. Proteins, DNA and other contaminants are removed in three rapid washing steps, and the bound RNA is then eluted in concentrated form. The entire procedure can be completed in 20 minutes, after tissue disruption. No phenol:chloroform extraction, protease digestion, or alcohol precipitation steps are required, making the procedure ideal for processing many samples. The resultant RNA is suitable for use in most common applications, including Northern blots, RT-PCR, nuclease protection assays, array probe labeling and in vitro translation. Ambion has incorporated the RNAqueous technology into a variety of products for specific applications.
