The RNAqueous-Midi Kit is designed for the purification of total RNA from 0.1 g to 0.5 g of tissue or 107–108 cells. In this kit, a luer-lock syringe filter houses the RNA binding glass fiber filter. Washing and elution solutions can be simply pushed through the filter using a standard 10 to 20 ml syringe (not included). No vacuum filtration steps are required; however, the syringe filter units also fit onto standard vacuum manifolds for processing several samples at once. The RNAqueous-Midi Kit provides enough reagents for 15 RNA purifications.
RNAqueous Technology RNAqueous kits can be used to purify total RNA from many different tissues and cells in a fast procedure without the use of phenol. Cells or tissue are disrupted in a guanidinium thiocyanate solution; this chaotropic agent effectively lyses cells and inactivates endogenous ribonucleases. The lysate is then diluted with an ethanol solution and applied to an RNA-binding glass fiber filter. Proteins, DNA and other contaminants are removed in three rapid washing steps, and the bound RNA is then eluted in concentrated form. The entire procedure can be completed in 20 minutes, after tissue disruption. No phenol:chloroform extraction, protease digestion, or alcohol precipitation steps are required, making the procedure ideal for processing many samples. The resultant RNA is suitable for use in most common applications, including Northern blots, RT-PCR, nuclease protection assays, array probe labeling and in vitro translation. Ambion has incorporated the RNAqueous technology into a variety of products for specific applications.
