Magnetic beads offer many benefits for isolating RNA from cells and viral RNA from cell-free samples. Ambion’s MagMAX™ technology binds RNA more efficiently than glass fiber filter methods, resulting in higher RNA yields. Furthermore, the RNA yields are more consistent, both from experiment to experiment, and over a broad range of sample sizes. Another important benefit of MagMAX over glass fiber filter-based methods is the elimination of filter clogging due to cellular particulates in samples. Since only a small volume of magnetic beads is needed for high efficiency binding, the bound RNA can be eluted in just 20–50 µl of elution buffer. This makes the MagMAX magnetic bead technology extremely useful for concentrating RNA from large, dilute samples.
The MagMAX-96 for Microarrays Total RNA Isolation Kit (patent pending) is designed for rapid, high throughput isolation of total RNA from mammalian cells and tissues in 96 well plates. The procedure employs the robust and reliable lysis/denaturant, TRI Reagent® and Ambion’s MagMAX magnetic bead-based RNA purification technology. The kit can also be used to process fewer than 96 samples at one time. The MagMAX-96 for Microarrays Kit is optimized both for use manually with multi-channel pipettors and for use with robotic liquid handlers (see Figure 2).

TRI Reagent lyses cells and facilitates sample homogenization while denaturing nucleases to maintain the integrity of RNA. By combining TRI Reagent and MagMAX technology, you get streamlined RNA purification, even in a high throughput format, without sacrificing RNA quantity or quality. To maximize efficiency and success, Ambion provides two alternative protocols for the kit. The “Spin” method has fewer steps, requires no DNase treatment, and allows for larger sample amounts to be processed. The “No Spin” method requires less sample handling and can easily be integrated with a robotic liquid handler.
The procedure is fast (< 1 hr), simple, and well suited for automation. The kit provides high yields of pure, intact RNA that can be used directly for quantitative reverse transcriptase PCR (qRT-PCR) and microarray analysis. Figure 3 shows RNA yield and quality data that illustrates the performance of the MagMAX-96 for Microarrays kit. In this experiment, highly intact RNA, with 28S/18S ribosomal RNA ratios of 1.1–1.4 was consistently purified from frozen mouse liver.
Successful gene expression studies require the use of high quality RNA. Figure 4 shows the high correlation between the method that Affymetrix recommends for producing RNA for use on their GeneChips® and the method that the MagMAX-96 for Microarrays kit employs. In addition, comparable Percent Present calls were obtained when using the MagMAX kit and the Affymetrix-recommended method (Figure 5).
