Designed to mimic endogenous precursor miRNAs

Anti-miR™ miRNA Inhibitors are chemically modified, single stranded nucleic acids designed to specifically bind to and inhibit endogenous microRNA (miRNA) molecules. These ready-to-use inhibitors can be introduced into cells using transfection or electroporation parameters similar to those used for siRNAs, and enable detailed study of miRNA biological effects (Figures 1,2).

Use of the Anti-miR miRNA Inhibitors enable miRNA functional analysis by down-regulation of miRNA activity. Specific experimental designs include:

  • miRNA target site identification and validation
  • Screening for miRNAs that regulate the expression of a gene
  • Screening for miRNAs that affect a cellular process

Cellular functions identified by the Anti-miR miRNA Inhibitors can be further investigated with the Pre-miR™ miRNA Precursor Molecules, which increase miRNA concentration for gain-of-function experiments.

Anti-miR™ let-7c miRNA Inhibitor Positive Control
Anti-miR hsa-let-7c miRNA Inhibitor provides a convenient, validated positive control for experiments using Anti-miR miRNA Inhibitors. Endogenous let-7 miRNA negatively regulates HMGA2, a ubiquitously expressed, nonhistone chromatin protein that modulates gene expression through changes in chromatin architecture. When transfected into human and mouse cells, Anti-miR hsa-let-7c miRNA Inhibitor blocks endogenous let-7c miRNA, resulting in increased levels of HMGA2 mRNA. Thus, Anti-miR hsa-let-7c miRNA Inhibitor activity can be monitored in human or mouse cells by qRT-PCR using a TaqMan® Gene Expression Assay to HGMA2 (Assay IDs: Hs00171569_m1 [Human], Mm00780304_sH [Mouse]). Anti-miR™ miRNA Inhibitor Negative Controls
Anti-miR™ Negative Control #1 is a random sequence Anti-miR molecule that has been extensively tested in human cell lines and tissues and validated to not produce identifiable effects on known miRNA function. Cy™3 and FAM™ dye-labeled Anti-miR Negative Controls are also available for monitoring transfection efficiency in transfection experiments using Anti-miR miRNA Inhibitors. The fluorescent label enables direct observation of the cellular uptake, distribution, and localization of the control. These dye-labeled controls are labeled at their 5' end and have the same oligonucleotide sequence as unlabeled Negative Control #1.

Anti-miR™ miRNA Inhibitor Libraries
The Anti-miR™ miRNA Inhibitor Library-Human V2 includes 327 miRNA inhibitors (0.25 nmol each) that correspond to 327 miRNAs in miRBase Sequence Database version 8.0.

The Anti-miR™ miRNA Inhibitor Library–Mouse V3 consists of 379 miRNA inhibitors corresponding to 379 mouse mature miRNAs cataloged in version 9.2 of the miRBase Sequence Database. This collection of miRNA inhibitors enables rapid study of miRNA function and can be used to simplify identification of miRNAs that interact with a particular target transcript. Each Anti-miR miRNA Inhibitor is supplied dried at 0.25 nmol in individual wells of a 96 well plate. This amount is sufficient for 50 transfections when used at 50 nM in 96 well plates.
