Silencer Pre-designed siRNAs, which are designed for maximum potency and specificity using a highly effective and extensively tested algorithm, are available for the human, mouse, and rat genomes. Each siRNA is synthesized to the highest quality standards and is provided with full sequence information. Furthermore, when you purchase three Silencer Pre-designed siRNAs to the same target, at least two of the siRNAs are guaranteed to reduce target mRNA levels by 70% or more.

Extensively Tested Algorithm
Silencer Pre-designed siRNAs are designed using a proprietary algorithm developed by Cenix BioScience and owned by Ambion (see siRNA Design: It's All in the Algorithm). Initial testing of the algorithm on 79 human genes indicated a 94% success rate. But additional testing was warranted to fully understand the algorithm's effectiveness on a genome-wide scale. Therefore, Cenix and Ambion directly measured the effectiveness of approximately 1,100 algorithm-designed siRNAs in the silencing of almost 400 endogenously expressed human transcripts (see lower figure at right). This comprehensive survey indicated that, on a per siRNA basis, more than 80% of the individual siRNAs showed >70% silencing of their target. The performance data clearly confirm the success of the Ambion siRNA selection process.

High Quality Synthesis from The RNA Company®
Ambion synthesizes and purifies each siRNA in state-of-the-art facilities to meet the highest quality standards. As part of our rigorous quality control procedures, each RNA oligonucleotide is analyzed by MALDI-TOF mass spectrometry and/or analytical HPLC. To provide the utmost in quality, Ambion also assesses each annealed siRNA by gel electrophoresis to confirm that the strands annealed properly. The result is premium quality siRNA that is purified and ready to use.

"In Vivo Ready" Silencer Pre-designed siRNAs—the highest quality siRNAs available anywhere—are subjected to that extra level of purification and testing required for the introduction of siRNAs into animals. After HPLC purification and annealing, each siRNA is further purified using a process that removes excess salt via a semi-permeable membrane. The result is highly pure siRNA with minimal salt content, suitable for in vivo applications. In vivo siRNAs are then sterile filtered and tested for the presence of endotoxin. At a concentration of 50 µM in deionized water, in vivo Ready siRNAs contain <0.6 mM Na⁺, <2.0 mM K⁺, and <0.1 mM Mg²⁺.
