Important Licensing Information

Ambion's custom siRNA service provides the premium quality siRNAs you need for your gene silencing research. Custom siRNAs can be readily ordered online by providing either the 19–21 base target sequence, or by providing the sense and antisense siRNA sequences. (For greater convenience, consider our Silencer® Validated and Silencer® Pre-designed siRNAs, with siRNAs already designed for all human, mouse, and rat targets in the RefSeq database.) Custom synthesized siRNAs are offered in various formats and amounts, and with many different modifications, including fluorescent labels. Premium Quality siRNA Synthesis
Each siRNA is synthesized in our state-of-the-art synthesis facility to meet the highest quality standards. Ambion's standard purification procedure, available for 21mer siRNAs, includes deprotection and column purification, and typically yields siRNA that is 90% pure (guaranteed >80% pure). For the most demanding applications, and for siRNAs of lengths other than 21 bp, HPLC purification is available, yielding siRNAs that are guaranteed >97% pure.

To ensure that the siRNAs are of highest quality, all RNA oligonucleotides are assessed by MALDI-TOF (matrix-assisted laser desorption ionization–time-of-flight) mass spectrometry, and the purity of all HPLC and PAGE purified oligonucleotides is assessed by analytical HPLC. Ambion also analyzes each annealed siRNA by gel electrophoresis to confirm that the strands annealed properly. The result is premium quality siRNA that is purified and ready to use.

In Vivo Ready siRNAs
"In Vivo Ready" custom synthesized siRNAs–the highest quality siRNAs available anywhere–are subjected to that extra level of purification and testing required for the introduction of siRNAs into animals. After HPLC purification and annealing, each siRNA is further purified using a process that removes excess salt via a semi-permeable membrane. The result is highly pure siRNA with minimal salt content, suitable for in vivo applications. In vivo siRNAs are then sterile filtered and tested for the presence of endotoxin. At a concentration of 50 µM in deionized water, in vivo Ready siRNAs contain <0.6 mM Na+, <2.0 mM K+, and <0.1 mM Mg2+.
