flashPAGE™ Fractionator
Ambion's flashPAGE Fractionator System (Figure 1) is the fastest, easiest, and most reliable method for isolating small nucleic acids shorter than 40 nt. This system has been designed and optimized for the isolation of mature miRNA (19–23 nt) from longer precursor molecules (Figure 2) and provides an efficient alternative to time-consuming polyacrylamide gel electrophoresis (PAGE) and subsequent gel elution (Figure 3), which have traditionally been required for isolation of small nucleic acids.

The flashPAGE Fractionator System includes four components: (1) the flashPAGE Fractionator unit, (2) pre-cast gels, (3) loading and running buffers, and (4) an optional flashPAGE Clean-up Kit for rapid concentration of fractionated samples.

To use the flashPAGE System, 1 to 100 µg nucleic acid (typically total RNA; the mirVana™ miRNA Isolation Kit can be used to prepare small RNA-containing RNA from cells and tissues) is loaded onto a flashPAGE Pre-Cast Gel inserted into the flashPAGE Fractionator unit (Figure 2). Samples are electrophoresed through the gel matrix at 75–80 V (constant voltage; 2–5 mA) for ~12 minutes. To concentrate the fractionated nucleic acid, the samples can be precipitated overnight in ethanol and sodium acetate, which results in >90% recovery. Use of the time-saving flashPAGE Clean-Up Kit, which features a 45 minute protocol, results in >70% yield. Extensive validation of the flashPAGE design confirms that greater than 95% of species longer than 40 nt are excluded from the small RNA/DNA fraction when the run is terminated with the included dye. Faithful representation of RNA is maintained.

The flashPAGE Fractionator comes with a 30 inch banana plug-type power cord, permitting use with any common laboratory power supply. Each instrument is covered by a 1 year warranty.

NOTE: Slight deviations exist between banana plugs, so an adapter may be needed to create a tight connection with some power supplies. Please contact your power supply manufacturer for the adapters.
